Looking to upgrade my 78 f150 400 with your saginaw conversion but I also plan on running hydro-boost and hydro-assist (eventually) so I'm looking at this pump. Can a fitting be used to cap/plug the unused return line until I need it?

Yes, you may plug one of the return ports. A plugged hose or tube cap with a clamp should do the job.
